Web of Life and Destiny is the centerpiece that turns all the Web Warrior allies into broken value engines. Mulligan aggressively for it. The only other cards worth keeping are Albert Moon and maybe Ingenuity. You will draw a lot of cards once Web of Life and Destiny is out, so don't worry about discarding other high value cards before you get it. Web of Life and Destiny is the glue that holds the entire Web Warrior package together so focus on seeing as many cards in your deck as quickly as you can until you get it.
Once you get Web of Life and Destiny out, you want to be playing a lot of Web Warriors every turn and Make the Call and Across the Spider-Verse lets you replay your best ones. They bypass Peter Parker's requirements, so you can use it to get him in play. They also let you bring back any high value Web Warriors you discarded in your search to get Web of Life and Destiny in play.
SP//dr is a core ally for any Web Warrior deck. The low cost along with the built in recursion means you're going to be spending 2 resources to thwart for 2, deal 1 damage, and draw a card every turn. Once you get this combo going, it will make the entire deck play smoother since she's a guaranteed +1 hand size every turn. It's well worth the 2 resource tax, especially once you get a Team-Building Exercise in play to help pay for her.
Spider-Man 2099 is the other key ally for this build. The key thing to note is that Web of Life and Destiny draws you a card when the ally leaves play not when they're defeated, so bringing an ally back to hand draws you a card immediately. Whereas SP//dr provides steady incremental value every turn, Spider-Man 2099 combos with Ghost-Spider and Hobie Brown to provide either repeated Swinging Silk Kick and Wallcrawl tutoring or repeated big damage hits.
Once Spider-Man 2099 gets low, you can either use his ability to bounce himself or recur him with Make the Call or Rapid Response. Rapid Response is preferable since Spider-Man 2099 is pretty expensive, so make sure you keep one around.
Otto Octavius is great for Silk once you get Organic Webbing in play. Organic Webbing isn't a tech upgrade, but a ready on a 2 cost ally is well worth it. Before that, his value is fairly limited, but a 2 cost Web Warrior early on is still good to buy another turn while still drawing you a card from Web of Life and Destiny.
Combining Spider Reflexes and Organic Webbing allows Silk to defend for up to 7 while still being ready during the hero phase. They provide a lot of consistency to your setup since you often don't want to defend with your Web Warriors. Eidetic Memory is a lot more situational, but it can very good especially in harder encounters.
